We are looking for a post-doctoral student for the development of a new project at the chemistry and biological interface funded by the National Agengy for Research (ANR � Labex). Such project is based on the collaboration between two laboratories located in Grenoble: the DCM (I2BM team) and the LAN -SCIB (CEA ). In this project, you will take part in a collaborative and multidisciplinary research program (organic chemistry, biochemistry, biology).
The work will consist in 1) The design and synthesis of DNA damage repair enzyme inhibitors. 2) The determination of their biological activity and inhibition efficiency in vitro and in cellulo. 3) The study of their mechanism of action.
The different steps of the research program will be:
- Inhibitors design. The compounds will be various cyclopeptide derivatives (synthesis, purification and characterisation) ;
- Structural studies (CD) ;
- Measurements of the biological activity and specificity of the synthetic compounds:
In vitro: tests using targeted enzyme (electrophoresis gel analysis, screening test using pro-fluorescent DNA probes).
In cellulo: cytotoxic study, measurement of the inhibitory efficiency in cellular culture.
- Study of the inhibitor interaction.
Organic Chemistry: 30% � Biochemistry/Biology: 70%.
Post-doctoral position for12 months, renewable 1 time, starting from March 2013.
Candidates profile:
You have a Phd in Biochemistry or Biological chemistry. You have an experience in the DNA repair and replication enzymology. Additional skill in cellular culture and (or) in bio-organic chemistry (nucleic acid and peptide synthesis) will be appreciated. Autonomy, rigor, curiosity and adaptability in a multidisciplinary team are qualities which will allow you to lead effectively this research program.
